Description & Applications
Description & origin
This fraction is generated via cation-exchange chromatography followed by gel filtration to isolate P-I and P-III snake venom metalloproteinases (SVMPs) from Bothrops jararaca. The process preserves catalytic zinc sites and maintains pro-domain integrity. SDS-PAGE and LC-MS confirm molecular masses between 23–60 kDa and minimal contamination by serine proteases.
Molecular targets
SVMPs cleave prothrombin and fibrinogen, activate Factor X, and degrade extracellular matrix components including laminin and collagen IV. Activity assays supplied in the COA quantify chromogenic substrate conversion and cell-based endothelial permeability shifts, enabling comparative analysis with mammalian MMPs.
Research applications
- Thrombosis modeling: Assess coagulation factor activation kinetics and evaluate antithrombotic candidates.
- Vascular permeability assays: Probe endothelial junction integrity under controlled proteolytic stress.
- Matrix metalloproteinase comparison: Benchmark SVMP-specific inhibitors and cross-reactivity against human MMPs.

Format & handling
Reconstitute 250 μg lyophilized powder with 250 μL sterile 20 mM Tris, 150 mM NaCl, pH 7.4. Include 5 mM CaCl₂ to stabilize catalytic domains. Maintain on ice during experimental setup.
Documentation
- COA: enzyme activity (U/mg), endotoxin (< 0.25 EU/mg), residual moisture (< 3%), and chromatographic purity.
- SDS: hazard classifications, PPE recommendations, spill response procedures.
Safety notes
Implement BSL-2 controls with splash protection. Neutralize unused material with chelating agents (EDTA) before disposal.
