This synthetic μ-conotoxin analog incorporates stabilizing substitutions (Cys-to-Pen) to maintain disulfide architecture while increasing oxidative resilience. Solid-phase peptide synthesis (Fmoc) with orthogonal cysteine protection yields controlled folding, followed by RP-HPLC purification. LC-MS verifies expected mass (2635.9 Da) and disulfide alignment.

Molecular targets

Selective inhibition of NaV1.7 (IC₅₀ 3.8 nM) with moderate activity on NaV1.4 (IC₅₀ 42 nM) and negligible effect on NaV1.5. The peptide slows recovery from inactivation, reducing repetitive firing in dorsal root ganglion neurons. COA includes voltage-dependence plots and protocol notes for step depolarization assays.
