Description & origin

The recombinant ω-conotoxin MVIIA is expressed in Pichia pastoris with secretory signal peptides enabling oxidative folding in the endoplasmic reticulum. Post-fermentation, the supernatant undergoes cation exchange and RP-HPLC purification. Final quality control includes mass spectrometry, analytical HPLC, and circular dichroism to confirm native-like folding.

Molecular targets

The peptide exhibits IC₅₀ 0.85 nM at CaV2.2 and 4.5 nM at CaV2.1. Whole-cell voltage-clamp assays on dorsal root ganglion neurons demonstrate > 90% reduction in high-voltage activated currents at 100 nM. The COA supplies inhibition curves and suggested protocols for cumulative dosing experiments.
