Operationalize Safe Venom Workflows

Use these checklists to standardize high-hazard reagent handling across teams and facilities.

Working with venom-derived reagents requires meticulous execution and documentation. The following protocols build a cohesive safety program that satisfies institutional biosafety committees and external auditors alike.

1. Pre-receipt preparation

  • Verify that the institutional biosafety committee (IBC) has approved the toxin class and proposed quantities.
  • Confirm that dedicated PPE (double gloves, splash-resistant lab coats, face shield) is stocked and inspected.
  • Calibrate temperature monitoring devices for cold-chain storage.

2. Receiving and accessioning

  1. Inspect outer packaging for damage; photograph any anomalies before opening.
  2. Review shipping temperature logs and compare against product specifications.
  3. Assign an internal accession number and record supplier lot, SKU, and expiration date in the toxin inventory system.

3. Storage controls

  • Segregate neurotoxins in locked, access-controlled freezers with continuous monitoring.
  • Apply tamper-evident seals to primary containers and record seal numbers in the inventory log.
  • Maintain secondary containment trays with absorbent liners for spill mitigation.

4. Reconstitution workflow

  1. Perform all manipulations in a Class II biosafety cabinet.
  2. Record the diluent, final concentration, technician initials, and time of reconstitution in the batch record.
  3. Prepare single-use aliquots to avoid repeated freeze–thaw cycles; label each aliquot with concentration, date, and accession number.

5. Experimental use and documentation

  • Use pre-printed worksheets to capture experiment ID, toxin aliquot ID, amount used, and waste disposition.
  • Require a second-person verification for calculations involving serial dilutions or unit conversions.
  • Update the inventory immediately after each use to maintain real-time traceability.

6. Spill response

  • Evacuate non-essential personnel and don splash protection.
  • Cover the spill with absorbent material, apply approved neutralizing solution (e.g., 10% bleach for peptide toxins), and allow appropriate contact time.
  • Collect waste into labeled hazardous containers and report the incident to EHS within one business day.

7. Waste disposal

  • Segregate sharp, liquid, and solid waste streams per local regulations.
  • Maintain manifests for third-party waste vendors, including chain-of-custody signatures.
  • Document final disposal in the inventory system to close out aliquot records.

8. Training and competency

  • Provide initial and annual refresher training that covers toxin properties, PPE, emergency response, and documentation requirements.
  • Use competency assessments (written quizzes and practical demonstrations) before granting independent access.
  • Retain training records for at least five years.
