Description & origin
This 38-residue peptide adapts an α-KTx scaffold with substitutions that reduce off-target Kv1.1 binding. Following SPPS synthesis, oxidative folding employs glutathione redox buffers to achieve the native disulfide pattern. The final product is purified by RP-HPLC and confirmed by LC-MS.
Molecular targets
Manual patch-clamp assays show IC₅₀ 0.85 nM against human Kv1.3 with > 50-fold selectivity over Kv1.1. The peptide exerts slow off-rate kinetics suited for sustained T-cell inhibition studies. COA includes kinetic constants derived from tsA201 cells expressing Kv1.3.
